CDK5在神经病理性疼痛中的作用

Abstract
细胞周期依赖性激酶5是一种在神经系统中高度表达的丝氨酸/苏氨酸激酶,其活性主要依赖于特异性激活蛋白p35或p39。近年来,大量研究证实CDK5在神经病理性疼痛的发生与维持中发挥关键调控作用。CDK5可影响突触可塑性,并通过磷酸化调控TRPV1、CaV3.2及CaV2.2等通道增强神经元兴奋性,并促进小胶质细胞和星形胶质细胞的活化,参与炎症放大反应。靶向CDK5的基因沉默策略与小分子抑制剂已表现出显著镇痛效果,提示其为神经病理性疼痛治疗的重要潜在靶点。
