肠道菌群与光化性角化病的因果关联:双向孟德尔随机化分析

Abstract
目的:采用两样本双向孟德尔随机化(MR)方法,探讨肠道菌群与光化性角化病(AK)之间的因果关系。方法:正向MR分析以GWAS数据中的肠道菌群为暴露变量、AK为结局变量;反向MR分析以AK为暴露变量,将正向分析中与AK显著相关的肠道菌群作为结局。因果效应主要通过逆方差加权(IVW)法估计,并结合多种MR方法,辅以异质性检验、多效性评估和敏感性分析,以验证结果的稳健性。结果:正向分析识别出12种风险菌群(如短芽孢杆菌目等)及15种保护菌群(如醋杆菌目等)(均P<0.05)。反向MR分析显示,AK仅对RC9sp900317925存在轻度正向调控(P<0.05)。敏感性分析结果支持上述结论的稳健性。结论:短芽孢杆菌目等12种肠道菌群可能与AK发病风险升高相关,而醋杆菌目等15种菌群可能对AK具有保护作用,为AK的预防和干预提供了潜在的微生态靶点。
