褪黑素在睡眠障碍治疗中的机制及药物相互作用研究进展

Abstract

褪黑素在调节昼夜节律以及在睡眠障碍治疗中具有重要作用,本文通过介绍睡眠的生理机制和作用机制,探讨了睡眠障碍的病因,并详细阐述了褪黑素的生理功能及其作用机制,表明褪黑素通过MT1受体抑制觉醒信号、MT2受体调节节律相位,并与GABA能系统协同作用促进深度睡眠。在药物治疗方面阐述了褪黑素能够改善患者的昼夜节律失调及苯二氮䓬类药物依赖等。其中褪黑素受体激动剂雷美替胺、阿戈美拉汀通过精准调控MT1、MT2受体及5-HT,展现出在抑郁失眠共病的应用潜力。表明了褪黑素与咖啡因、β受体阻滞剂等存在拮抗作用以此引出对褪黑素的安全性进行探讨,在最后提出对未来研究方向的建议。
